Estradiol and genistein exaggerate cardiac hypertrophy in ovariectomized AhR knockout mice with different effects on gene expression of hypertrophy markers.

Cardiac hypertrophy was shown as a manifestation of AhR knockout (AhRKO) mice suggesting an important role of AhR in heart homeostasis and development. In addition, AhR – estrogen receptor (ER) crosstalk has been studied in different cell lines indicating AhR-dependently functional pathways of estrogenic compounds in general and AhR-correlated estrogen-based differences in heart morphology and functions in particular. This study identifed effects of estradiol (E2) and genistein (Gen) on hypertrophic heart of AhRKO mice. Ovariectomized (ovx) AhRKO mice showed heart hypertrophy. Oral treatment with both E2 and Gen increased the hypertrophy levels. The increase tendency in was: Gen AhRKO > E2 AhRKO > control AhRKO > wildtype control. Gene expression of hypertrophy makers, alpha- and beta- myosin heavy chains was increased by E2, whereas Gen treatment led to lower mRNA levels of atrial natriuretic factor. Conclusions:.E2 and Gen might exaggerate AhRKO-induced cardiac hypertrophy with different effects on cardiac gene expression.
