Objectives: Despite the presence of both known types of estrogen receptors (ER) in the heart, only limited data about genes involved in the actions of E2 in the heart under physiological conditions are available. Thus, studies on the effects of E2 on gene- and protein expression in the myocard are required. To decipher the role of either ER, experiments have to be done in both, wildtype and in ER - knockout animals. Since only ER knockout mice but not rats are available, the established protocols for treatment of ovariectomized (ovx) rats have to be validated in mice. Therefore, we performed a 3 month study with ovx mice in order to compare the effects of E2 on the target organs uterus, bone and heart.

Methods: Mice were ovx at 2 months of age. Two groups of 20 mice each were fed either with soy free pelleted chow containing E2 or soy free diet as control. After 3 months mice were sacrificed. Blood was collected for serum analysis, heart and uterus were excised and weighed. The tibia was collected for bone mineral density (BMD) measurement by computer-assisted tomography (CT).

Results: Mean E2 concentration in the serum of steroid treated mice resulted in physiological levels. In comparison to the control group, body weight (BW) of E2 treated mice decreased while the uterus weight increased leading to increase of uterus/BW ratio. Heart weight/BW ratio was raised by E2. Trabecular BMD in the metaphysis of the tibia from E2 treated mice was significantly increased compared to control mice. A microarray analysis of heart RNA from both groups is currently in progress.

Conclusions: Long term application of E2 via pelleted food resulted in physiological hormone levels. In mice, the target organs uterus, bone and heart are affected by E2 in similar manner as reported previously for rats. Thus, our treatment protocol proved to be valid to study the molecular mechanism of ER actions and their ligands in knockout mice

Mặc dù viêc xác định vai trò của gene và protein thường được tiến hành trên cả hai đối tượng mẫu động vật bình thường và động vật bất hoạt gene nhưng hiện nay các thí nghiệm sử dụng động vật bất hoạt gene cho nghiên cứu vai trò của estrogen receptor được tiến hành trên chuột nhắt (mice) trong khi nghiên cứu ở trạng thái sinh lý mới được tiến hành trên chuột (rat). Do các kết quả sử dụng hai đối tượng này không phải lúc nào cũng ứng dụng được cho nhau nên thí nghiệm này được tiến hành với mục đích thiết lập mẫu nghiên cứu sinh lý tim (dưới tác động của estrogen) trên chuột nhắt làm cơ sở cho việc sử dụng chuột mang gene bất hoạt qua kiểm tra các chỉ tiêu mật độ xương chày với kỹ thuật CT scan, khối lượng và kích thước tử cung, mRNA và IGF-1 (tử cung, gan và tim) bằng real time RT-PCR cũng như nồng độ estradiol huyết thanh dùng radioimmunoassay.
